Administration of Ginkgo Biloba Extract (Egb761) Alone and in Combination with FK506 Promotes Liver Regeneration in a Rat Model of Partial Hepatectomy

Background: Free radical damage is known to occur during liver regeneration. EGb761 is used for its antioxidant feature by action of its ability to scavenge free radicals. FK506 has been used widely as an immunosuppressant which stimulates hepatocyte proliferation in partial hepatectomy.

Aims: To explore whether ginkgo biloba extract (EGb761) enhances liver regeneration after hepatectomy liver regeneration in rats. Therefore, we have investigated the effects of EGb761 on liver regenerative process, alone and together with FK506.

Study Design: Animal experimentation.

Methods: Seventy-five Wistar albino rats, weighing 340.08±11.66 were randomly divided into five experimental groups as a sham, control, FK506, EGb761, and a combination of these drugs.

According to the group of study, rats received 0.1mg/kg/day of FK506 and 25mg/kg/day of EGb761 three times by intraperitoneal route. After of pre-therapy, two-thirds hepatectomy was performed according to the Higgins and Anderson technique to all rats. Forty-eight hours after hepatectomy, 53 surviving rats were sacrified; there maining regenerated liver was entirely resected, weighed and sampled for mitotic index on hematoxylin-eosin staining. Also, serum and plasma samples were taken for analyses of thymidine kinase, oxidative stres and other parameters during final operation. Liver regeneration calculated by Child’s formula. Data were statistically analyzed by ANOVA with significance level of 5% (p<0.05).

Results: Rats receiving EGb761 showed statistically significant higher levels of liver regeneration when compared to FK506 and its combination (p<0.01). Similar trend was found in EGb761 (p<0.005) and FK506 (p<0.05) when compare to control and sham group for thymidine kinase and mitotic index. In addition, liver regeneration percentage was revealed higher significant in EGb761 alone compare to FK506 group (p<0.01). Myeloperoxidase and malondialdehyde levels were shown significant correlation between EGb761 and FK506 groups (even at lower levels in EGb761 group) (p<0.001).

Conclusion: EGb761 counteracts oxidative stres and decreases liver damage. So we suggested that, EGb761 pretreatment have reduced liver damage and stimulated liver regeneration after in rat partial hepatectomy model through anti-inflammatory and anti-oxidative effects.
